Fiber size diameters of immunohistochemical fiber types were measured in eight confocal fields of view at 10 × magnification. Fiber types were identified using a combination of staining patterns of the A4.74 and 6H1 antibodies [16]. Fibers were determined to be type MyHC-IIA with intense A4.74 staining and without 6H1 colocalization (n = 41), or MyHC-IIA/IIX hybrid fibers with intensive A4.74 positivity colocalizing with 6H1 staining (n = 20). MyHC-IIA and MyHC-IIA/IIX fibers were significantly smaller in diameter and more infrequent than MyHC-IIX fibers (n = 172) (weak A4.74 positivity colocalizing with 6H1 staining) and type I fibers (n = 112), with negative A4.74 and 6H1 staining but visualized and measured in the DAPI channel.
