Temporal lobe homogenates probed with AT8 and isoform-specific antibodies, RD3 and RD4. In sarkosyl-insoluble preparations, AT8-positive a and RD4-positive c bands were detectable between 30 and 35KDa in aged monkeys (asterisk 32, 35, 36 and 36 years-old) but not in young controls (7 years-old). This age associated increase is neither evident in Tris-soluble preparations probed by AT8 b, nor in sarkosyl-insoluble preparations probed with RD3 d. Higher molecular complex at 50–70 kDa, seen in human brain with tau deposits, were not detectable in either sarkosyl-insoluble preparations or tris-soluble preparations
