Condition-specific immunoreactivity of the hSOD and C4F6 antibodies to purified recombinant hSOD1. (a) 1.5 μg of the indicated recombinant hSOD1 protein was spotted onto a nitrocellulose membrane. All membranes were treated with 4% paraformaldehyde followed by either no treatment, 6 M guanidine hydrochloride (Para/GdnHCl), or steamed in citrate buffer (Para/Cit.Buff.) as described in Methods. Membranes were then blotted with the hSOD (a) and C4F6 (c) antibodies. (b and d) The intensities of the spots were quantified using an Odyssy Imaging System with the values normalized to the WT signal, for the hSOD blots, or to the G93A signal, for the C4F6 blots; the data from 3 independent experiments are graphed (mean ratio ± S.E.
