Aβ deposition patterns in the brain of AD, MCI, NDAN, and Aged control individuals. (a) The Aβ aggregation profile in the cingulate cortex of a typical AD case was compared to the one present in MCI and NDAN individuals. Slides were stained with an anti-Aβ (4G8) antibody, thioflavin S (ThS) and anti-hyperphosphorylated tau protein (AT8) antibody. Brain slices from an aged non-demented control did not show any AD-associated characteristic (right panels). The scale bar corresponds to 100 μm. (b and c) Aβ levels in these brains were evaluated by serial extraction and ELISA as described in Methods. The quantity of Aβ40 (b) and Aβ42 (c) was measured using ELISA kits specifically designed to identify these peptides. PBS, Phosphate Buffer Saline; SDS, Sodium Dodecyl Sulfate; FA, Formic Acid. The values were expressed as means ± SEM of the different brains used. Samples were measured in duplicates.
